Utilizzo di linfociti T ingegnerizzati nell’ipercolesterolemia familiare

L’ipercolesterolemia familiare (FH) è un malattia genetica caratterizzata da livelli molto elevati di colesterolo Ldl che può portare, in assenza di trattamento, a malattia coronarica e mortalità precoce. Attualmente l’efficacia della terapia farmacologica è limitata da pesanti effetti collaterali, perché per controllare la malattia occorrono dosaggi molto elevati. Studi recenti suggeriscono che la patologia si associ a un’attivazione precoce e robusta della risposta immunitaria, come conseguenza della perdita di tolleranza e aumento di reattività alle Ldl. Obiettivo di questo progetto è valutare le potenzialità terapeutiche di un approccio basato su una versione ingegnerizzata di una specifica popolazione di cellule immunitarie, i linfociti T helper. Così manipolate, queste cellule sarebbero in grado di esprimere in maniera costante sulla loro superficie una particolare proteina, il recettore CCR2 (CCR2-Treg), che le renderebbe capaci di essere reclutate in modo selettivo a livello delle placche aterosclerotiche tipiche di questa patologia. In questa sede, le cellule manipolate potrebbero interferire in maniera positiva con l'eccessiva risposta immuno-infiammatoria e migliorare così il decorso della patologia.
